My sister got her results 2 days ago, and she's now got to wait for her enrolment at the Jane Austen College for sixth form.

Yes you have to defeat him once to trigger. When I converted my pesticides I got plenty of 2 of them & none of the futuristic harpoon which like you I needed to start it off.

I've already defeated him 3 times for the challenge. The plan is to defeat him as many times as I can with the harpoons I have, wait for the timer to reset, and then defeat him again as many times as I can. Rinse and repeat.

I ran out of harpoons again, just need to build up. But so far I have defeated demon stewie 8/9 times, so tomorrow night I should have the Gorge-ous Bike.

I got lucky and got it the day it launched thanks to all the pesticide I had it also got me the action movie set

I've been crazy busy with work and missed out on a couple of the timed challenges... I wont get the current bike... which means I wont get the 5th bike either... FML!

I did the same.  Nathan was required for 22 hours (now I find out it's another 24 after that..) so I held off until the Harpoon challenges were done.  Lois held off on the DNA examination as well.  Didn't want to lock up my producers doing long quests.

So nobody can get the final 2 vehicles? Latch Stewie spawns every 4 hours, that's 6 a day for 4 days. You can only get 24 total without missing any, but need to get 25. Not even clams can help you

Sorry, I wouldn't know, I believe Quagmire has nothing to do with it.  I saw a new update and the quests just showed up.  I finished every mission as well, but the two that showed up last night for Bryan Cranston and George Takei.
